目的 证实大鼠坐骨神经上ATP受体P2y的分布。 方法 梨状肌下缘以远 5mm处切取长 5mm的坐骨神经 ,同时切取主动脉 3mm作阳性对照 ;组织总mRNA提取 ,RT PCRcDNA第一链合成 ,反应所得产物于 2 %琼脂糖凝胶电泳 ,1μg/ml溴化乙啶染色 (EB) ,观察 ,照相。 结果 在大鼠坐骨神经上有 3种ATP受体亚型表达 (P2y2、P2y4和P2y6mRNA表达 )。 结论 大鼠坐骨神经上有 3种ATP受体亚型分布 ,细胞外ATP可能通过此三种受体或其中之一完成轴突诱向作用 ,P2y受体与神经损伤修复有关
Objective To confirm the distribution of ATP receptor P2y on rat sciatic nerve. Methods The sciatic nerve 5 mm long was cut away from the lower edge of the piriformis by 5mm and the aorta was cut at 3mm for positive control. Tissue mRNA was extracted and synthesized by first-strand RTcccDNA. The reaction products were separated on 2% agarose gel electrophoresis and 1μg / ml Ethidium bromide staining (EB), observation, photography. Results There were 3 ATP receptor subtypes (P2y2, P2y4 and P2y6 mRNA expression) in rat sciatic nerve. Conclusion There are three types of ATP receptor subtypes on the sciatic nerve in rats. Extracellular ATP may induce axon induction through one or both of these three receptors. P2y receptors are involved in the repair of nerve injury
