本文采用快速印迹杂交(quickblot)和延续转录试验(run-oftranscriptionassay)两种特异mRNA检测技术,在经β-萘黄酮(β-NF)诱导的人羊膜上皮细胞FL系中,观察了茶多酚对人羊膜上皮细胞FL系CYP1A1mRNA水平和转录的影响,结果表明,细胞中CYP1A1mRNA的含量和细胞核CYP1A1转录水平均比空白对照组增加,茶多酚可明显抑制其基础表达及β-NF的诱导作用。因此,我们认为茶多酚预防肿瘤的作用还可能与它影响细胞CYP1A1基因表达,从而影响CYP1A1同功酶活性有关。
In this paper, two specific mRNA detection techniques, rapid blot and run-oftranscription assay, were used to observe the polyphenols in the human amniotic epithelial cell FL induced by β-naphtlavone (β-NF). The effect of CYP1A1 mRNA level and transcription on human amniotic epithelial cells FL line CYP1A1 mRNA levels and transcription levels in the cell nucleus CYP1A1 were increased compared to the blank control group. Tea polyphenols significantly inhibited the expression of CYP1A1 mRNA and the induction of β-NF. . Therefore, we believe that the role of tea polyphenols in preventing tumors may also be related to its effect on cell CYP1A1 gene expression, thereby affecting the CYP1A1 isoenzyme activity.
