目的 探讨载脂蛋白J (apoJ)在大鼠脑梗死急性期的作用及其可能的机制。 方法 清洁级SD雄性成年大鼠 84只 ,随机分为正常对照组、假手术组、脑梗死后 6h组、2 4h组、3d组、5d组和 7d组。用线栓法制备永久性局灶脑梗死模型 ,动物处死后测脑含水量 ,并检测apoJ及补体C3mRNA的表达值。 结果 对照组大鼠脑内有apoJmRNA表达 ;各模型组缺血侧ApoJmRNA表达增高。ApoJmRNA表达值与脑含水量呈负相关 (r=- 0 .2 12 ,P <0 .0 5 ) ,与补体C3mRNA表达水平呈正相关 (r =0 .5 0 9,P <0 .0 1)。结论 大鼠脑梗死后apoJ表达增高 ,3d达高峰。ApoJ可能有保护缺血脑组织的作用。
Objective To investigate the role of apolipoprotein J in acute stage of cerebral infarction in rats and its possible mechanism. Methods Totally 84 SD adult male SD rats were randomly divided into normal control group, sham operation group, 6h group after cerebral infarction, 24h group, 3d group, 5d group and 7d group. The model of permanent focal cerebral infarction was prepared by thread method. The brain water content was measured after sacrificed and the expression of apoJ and complement C3 mRNA was detected. Results There was apoJ mRNA expression in the rat brain in control group. The expression of ApoJ mRNA in ischemic side increased in each model group. ApoJ mRNA expression was negatively correlated with brain water content (r = - 0.22, P <0.05), and positively correlated with complement C3 mRNA expression (r = 0.509, P <0.01) . Conclusion ApoJ expression increased after cerebral infarction in rats and peaked at 3d. ApoJ may protect ischemic brain tissue.
