本文研究各种眼镜蛇科、响尾蛇科和蝰蛇科蛇素在体外.体内对 B16F10黑色素瘤和体内对软骨肉瘤的细胞毒性。细胞毒性的测定在体外采用瘤细胞培养法:把含10~5黑色素瘤细胞的悬液1ml 置于3厘米的佩特里细菌培养皿中,每个培养皿加含6微克天然蛇毒的磷酸缓冲液(0.01M,PH7.0)10微升,培养24小时。用倒置相差显微镜连续观察瘤细胞,通过计数受损细胞(失去突起和与基质分离)的数量来决定细胞毒性的大小。体内试验通过在 C_(57)黑小鼠皮下或尾静脉注射瘤细胞与蛇毒的混悬液来进行,对照组只注射瘤细胞,2——3周后观察注射部位及肺部的肿瘤。
This article investigates the cytotoxicity of various Cobra, Rattlesnake, and Vipera in vitro on B16F10 melanoma and in vivo on chondrosarcoma. Cytotoxicity was measured in vitro using a tumor cell culture method: 1 ml of a suspension containing 10 to 5 melanoma cells was placed in a 3 cm petri dish, and 6 μg of natural venom phosphate buffer was added to each dish. 10 μl of liquid (0.01 M, pH 7.0) was cultured for 24 hours. The tumor cells were continuously observed using an inverted phase contrast microscope, and the number of cytotoxic cells was determined by counting the number of damaged cells (loss of protrusion and separation from the matrix). The in vivo test was performed by subcutaneous or caudal vein injections of tumor cells and venom suspensions in C57 mice. In the control group, tumor cells were injected. After 2 to 3 weeks, tumors at the injection site and in the lungs were observed.
