目的探讨加味生脉散对鼻咽癌裸鼠移植瘤放疗增敏作用及相关机制。方法构建带绿色荧光蛋白的鼻咽癌细胞HONE1稳定株,并接种裸鼠成瘤,实验分为对照组、药物组、放疗组、药物+放疗组,通过测量肿瘤体积及计算抑瘤率,观察药物加味生脉散的放疗增敏作用;荧光定量PCR及Western-blot检测瘤组织MDR1基因的表达差异。结果与对照组相比,放疗组及药物+放疗组的肿瘤抑制率均明显升高(P<0.05),其中以药物+放疗组最明显(P<0.01);MDR1的mRNA及蛋白的表达水平在单纯放疗组与药物+放疗组之间差异明显(P<0.01)。结论加味生脉散对鼻咽癌的放疗具有增敏作用,其增敏机制可能与抑制MDR1基因的表达相关。
Objective To investigate the radiosensitizing effect of Jiawei Shengmaisan on radiotherapy of nasopharyngeal carcinoma in nude mice and its related mechanism. Methods Stable HONE1 cells with green fluorescent protein (EGFP) were constructed and inoculated into nude mice. The experiment was divided into control group, drug group, radiotherapy group and drug + radiotherapy group. The tumor volume was measured and the inhibition rate was calculated The drug-treated Shengmai Powder has the radiosensitizing effect. The difference of MDR1 gene expression was detected by quantitative PCR and Western-blot. Results Compared with the control group, the tumor inhibition rates of the radiotherapy group and the drug + radiotherapy group were significantly increased (P <0.05), especially in the drug + radiotherapy group (P <0.01). The mRNA and protein expression of MDR1 The difference between the radiotherapy group and the radiotherapy + radiotherapy group was significant (P <0.01). Conclusion Jiawei Shengmaisan can enhance the radiosensitivity of nasopharyngeal carcinoma. The mechanism of its sensitization may be related to the inhibition of MDR1 gene expression.
