目的:探讨姜黄素对缺氧复氧的乳鼠心肌细胞是否具有保护作用,这种保护作用是否与热休克蛋白70(HSP70)表达有关。方法:原代培养乳鼠心肌细胞,MTT法测IC50确定姜黄素浓度,建立缺氧复氧模型,分别测定各组细胞乳酸脱氢酶(LDH)活性、丙二醛(MDA)含量,缺氧/复氧细胞凋亡与坏死,并用免疫组织化学检测HSP70表达。结果:缺氧/复氧模型预先加姜黄素组较I/R组LDH活性与MDA含量下降,HSP70表达增加,细胞凋亡与坏死少。结论:姜黄素对体外培养的缺氧复氧心肌细胞具有保护和抗凋亡作用,姜黄素对缺氧复氧心肌细胞保护作用可能与HSP70表达增加有关。
Objective: To investigate whether curcumin can protect cardiomyocytes from hypoxia-reoxygenation injury and whether this protective effect is related to the expression of heat shock protein 70 (HSP70). Methods: Primary cultured neonatal rat cardiomyocytes were cultured in vitro. The concentration of curcumin was determined by MTT assay and the hypoxia-reoxygenation model was established. The activities of lactate dehydrogenase (LDH), malondialdehyde (MDA) / Reoxygenation apoptosis and necrosis, and immunohistochemical detection of HSP70 expression. Results: The hypoxia / reoxygenation model preaddition curcumin group compared with I / R group LDH activity and MDA content decreased, HSP70 expression increased, apoptosis and necrosis less. CONCLUSION: Curcumin has protective and anti-apoptotic effects on hypoxia-reoxygenation cardiomyocytes cultured in vitro. The protective effect of curcumin on hypoxia-reoxygenation cardiomyocytes may be related to the increase of HSP70 expression.
