目的酸应激对胃癌SGC7901细胞自噬强度的影响。方法选择人胃癌细胞系SGC7901作为研究细胞,用调节到pH 6.5或pH 7.4的完全培养基处理胃癌SGC7901细胞。采用免疫荧光检测酸应激时胃癌细胞系SGC7901中自噬体的形成情况,采用Western-Blot检测自噬相关蛋白LC3-Ⅱ的表达情况。结果激光共聚焦电镜下观察到酸应激处理的胃癌SGC7901细胞荧光呈颗粒状数较对照组细胞内荧光颗粒数显著增多。酸性完全培养基处理的胃癌SGC7901细胞较对照组SGC7901细胞LC3-Ⅱ生成增多,p62表达减少。结论酸应激可以引起胃癌SGC7901细胞自噬增强。
Objective To investigate the effect of acid stress on autophagy of gastric cancer cell line SGC7901. Methods Human gastric cancer cell line SGC7901 was selected as a research cell and gastric cancer SGC7901 cells were treated with complete media adjusted to pH 6.5 or pH 7.4. The formation of autophagosomes in gastric cancer cell line SGC7901 was detected by immunofluorescence. The expression of autophagy-related protein LC3-Ⅱ was detected by Western-Blot. Results It was observed by laser scanning confocal microscopy that the number of fluorescent particles in SGC7901 cells treated with acid stress was significantly higher than that in control cells. SGC7901 cells treated with acid complete medium increased LC3-Ⅱ production and decreased p62 expression in SGC7901 cells. Conclusion Acid stress can induce autophagy in gastric cancer SGC7901 cells.
