以“郑红 5”等 6个甘薯品种为材料 ,通过离体培养诱导体细胞胚胎发生与植株再生。利用 RAPD分子标记方法 ,在 DNA水平上分析甘薯品种间遗传差异以及体细胞胚胎无性系的遗传变异。筛选了 45个随机引物 ,其中 7个引物 PCR扩增后在品种间和体细胞胚胎无性系中表现 RAPD多态性 ,并在相应胚性愈伤组织中检测到少数 RAPD变异位点。证明利用 RAPD分子标记方法鉴定甘薯体细胞胚胎发生遗传变异的可行性 ,以及为通过离体培养条件调控 (增加或减少 )甘薯体细胞胚胎发生中的遗传变异提供了可靠依据
Six sweet potato varieties such as “Zheng Hong 5” were used as materials to induce somatic embryogenesis and plant regeneration through in vitro culture. RAPD molecular markers were used to analyze the genetic differences between sweet potato varieties and the genetic variation of somatic embryos clones at the DNA level. 45 random primers were screened. Among them, 7 primers amplified RAPD polymorphism among varieties and somatic embryos clones after PCR amplification, and a few RAPD mutation sites were detected in the corresponding embryogenic callus. The feasibility of using RAPD markers to identify the genetic variation in somatic embryos of sweet potato and provide a reliable basis for regulating (increasing or decreasing) the genetic variation in somatic embryogenesis of sweet potato by in vitro culture conditions
