目的:观察复方贞术调脂方对长期高脂饮食致非酒精性脂肪肝(NAFLD)小鼠的防治作用及肝脏内质网应激(ERS)的调节作用。方法:采用高脂纯化配方饲料喂养建立NAFLD小鼠模型,同时防治性给予复方贞术调脂方20 w,通过观察血浆、肝脏的总胆固醇(TC)和甘油三酯(TG)水平、肝组织病理形态学以及脂质代谢和ERS相关基因的表达,考察复方贞术调脂方对NAFLD的防治作用及机制。结果:与正常对照组比较,模型组血浆及肝脏组织中TC、TG含量及肝组织PERK mRNA表达显著升高(P<0.05);与模型组比较,复方贞术调脂方低剂量组血浆及肝组织中TC、TG含量均显著降低(P<0.05),肝组织XBP-1、PERK、SREBP-1c mRNA表达显著下调(P<0.05)。结论:复方贞术调脂方能显著缓解长期高脂饮食诱导的小鼠NAFLD形成,调节肝脏ERS可能是其作用途径之一。
Objective: To observe the preventive and therapeutic effects of Fufang Zhenzhu Tiaozhi Recipe on long-term high-fat diet induced non-alcoholic fatty liver disease (NAFLD) mice and the regulation of liver endoplasmic reticulum stress (ERS). Methods: NAFLD mice model was established by feeding with high-fat purified formula. At the same time, anti-hyperlipidemia prescription was given for 20 weeks. The levels of total cholesterol (TC) and triglyceride (TG) Pathological morphology and lipid metabolism and expression of ERS-related genes, to investigate the compound virginity Tiaozhi prescription on the prevention and treatment of NAFLD and its mechanism. Results: Compared with the normal control group, the contents of TC and TG and the expression of PERK mRNA in liver and liver were significantly increased in the model group (P <0.05). Compared with the model group, The levels of TC and TG in liver tissue were significantly decreased (P <0.05), and the expressions of XBP-1, PERK and SREBP-1c in liver tissue were significantly decreased (P <0.05). Conclusion: FZF can significantly alleviate the formation of NAFLD induced by long-term high-fat diet in mice, and regulation of hepatic ERS may be one of its mechanisms.
