目的探讨富含亮氨酸重复序列的G蛋白偶联受体(LGR5)在小鼠结肠癌模型中的表达特点。方法选取80只6~10周龄C57/BL6小鼠,采用腹腔注射氧化偶氮甲烷法建立小鼠结肠癌模型,以生理盐水注射组作为对照,采用RT-PCR和免疫组化方法检测LGR5 m RNA及蛋白在小鼠结肠癌组织中的表达特点。结果接受氧化偶氮甲烷注射的40只小鼠中有30只成功诱发结肠癌,其中24只结肠癌中LGR5 m RNA和蛋白表达水平显著升高;与LGR5阴性细胞相比,LGR5阳性细胞中的Ki67表达水平明显升高。结论 LGR5在结肠癌组织中表达水平显著升高,可能在结肠癌的发生发展中起到了重要作用。
Objective To investigate the expression of leucine-rich G protein-coupled receptor (LGR5) in mouse colon cancer model. Methods Eighty C57 / BL6 mice aged 6-10 weeks were selected. The mouse model of colon cancer was established by intraperitoneal injection of azoxymethane. The normal saline injection group was used as a control. The expression of LGR5 was detected by RT-PCR and immunohistochemistry Expression of RNA and protein in mouse colon cancer. Results Thirty of the 40 mice injected with azoxymethane successfully induced colon cancer, and the expression of LGR5 m RNA and protein in 24 colon cancer tissues was significantly increased. Compared with LGR5-negative cells, LGR5-positive cells Ki67 expression was significantly increased. Conclusion The expression of LGR5 in colon cancer tissues is significantly increased, which may play an important role in the development of colon cancer.
