目的:探讨超滤膜技术分离纯化掌叶大黄多糖的可行性工艺。方法:采用正交设计法优选超滤前离心条件;以超滤压力、料液温度及料液浓度为考察因素,以粗多糖得率和含量为考核指标,均匀设计优选超滤工艺参数。结果:最佳超滤条件为药液以3 000 r.min-1离心处理12 min,用分子截留量10万的膜,超滤压力为0.05~0.09 MPa,药液温度40℃,药材与提取液质量比1∶10~1∶30,pH 6.0,超滤膜先用tween-80(5 g.L-1)处理10 min。结论:优化所得超滤工艺与未超滤相比有较高的多糖得率和含量。超滤技术可用于纯化、富集掌叶大黄多糖,具有产业化开发前景。
Objective: To investigate the feasibility of separation and purification of polysaccharide from Rheum palmatum by ultrafiltration membrane technology. Methods: The orthogonal design method was adopted to optimize the centrifugation conditions before the ultrafiltration. Taking the ultrafiltration pressure, the temperature of the feed liquid and the concentration of the feed liquid as the investigation factors, the yield and content of the crude polysaccharide were taken as the assessment indexes to optimize the UF process parameters. Results: The optimal ultrafiltration conditions were centrifugation at 3000 r.min-1 for 12 min, ultrafiltration pressure of 0.05 ~ 0.09 MPa, liquid temperature of 40 ℃, Liquid mass ratio 1:10 ~ 1:30, pH 6.0, the first ultrafiltration membrane with tween-80 (5 gL-1) for 10 min. Conclusion: The optimized ultrafiltration process has a higher yield and content of polysaccharides than non-ultrafiltration. Ultrafiltration technology can be used for purification, enrichment palm leaf rhubarb polysaccharides, with industrial development prospects.
