采用Southern吸印杂交技术,对人乳头瘤病毒16型(HPV-16)在人宫颈癌活检组织中存在的状况进行了研究。结果提示:①存人宫颈癌组织中存在着HPV-16 DNA序列,并且HPV-16 DNA是整合在肿瘤细胞的基因组中(有的也带有游离的片段)整合的位点是随机的;②不同的人宫颈癌组织HPV-16DNA的含量不同,酶切片段的大小也不同;③HPV-16具有诱导NIH/3T3细胞发生转化的功能,病毒基因亦是整合在细胞染色体DNA中。
The Southern blotting hybridization technique was used to study the existence of human papillomavirus type 16 (HPV-16) in human cervical cancer biopsies. The results suggest that: (1) The HPV-16 DNA sequence exists in human cervical cancer tissues, and HPV-16 DNA is integrated into the genome of tumor cells (some also have free fragments); the site of integration is random; Different human cervical cancer HPV-16DNA content, digested fragments of different sizes; ③ HPV-16 has the function of inducing NIH / 3T3 cell transformation, the virus gene is also integrated in the cell chromosomal DNA.
