目的 评价人透明带空腔冻存精子的可行性。方法 本文用微注射针将体外受精治疗过程中的未成熟卵胞浆抽吸干净 ,每个透明带空腔内注入 10条运动精子。在含甘油及HSA等的HTF -HEPEs冻存液中 ,将精子冻存于液氮中。解冻后检测精子活动率和存活率 ,并用显微授精方法将冷冻精子注射入废弃的人卵母细胞内观察冻存精子的受精能力。结果 射精液精子冻融后的活动率为 63 .4%± 18.1% ,存活率为 70 .7%± 14.6% ;单精子注入卵母细胞浆内 ,能获得 61.2 %± 10 .7%的受精率 ;6例附睾精子冻融后的活动率和存活率分别为 5 2 .0 %±7.5 %和 65 .0 %± 5 .5 % ,显微授精后的受精率为 5 3 .8%± 8.6%。结论 人透明带空腔是一种有效的冻存稀少精子的载体
Objective To evaluate the feasibility of cryopreservation of sperm in zona pellucida. Methods In this study, immature oocyte cytoplasm during IVF treatment was aspirated cleanly with microinjection needle, and 10 motile spermatozoa were injected into the cavity of each zona pellucida. In HTF-HEPEs cryopreservation solution containing glycerol and HSA, sperm are frozen in liquid nitrogen. After thawing, the sperm motility and survival rate were measured, and frozen sperm was injected into the abandoned human oocyte by micro-insemination to observe the fertilization ability of cryopreserved sperm. Results The sperm motility after elution was 63.4% ± 18.1% and the survival rate was 70.7% ± 14.6%. The single spermatozoa were injected into the oocyte plasma to obtain 61.2% ± 10.7% of fertilization The activities and survival rates of sperm in 6 cases of epididymis were 52.0% ± 7.5% and 65.0% ± 5.5%, respectively. The fertilization rate after sperm fertilization was 53.8% ± 8.6%. Conclusion Human zona pellucida cavity is an effective cryopreserved rare sperm carrier
