目的建立和评价血清中对氧磷酯解酶1(PON1)的自动化检测方法。方法在Hitachi7600上建立自动化测定血清PON1参数。对该方法的重复性、线性、干扰分析进行评估,并对正常人血清PON1含量进行分析。结果低、中、高三个浓度PON1的总变异分别为6.5%、4.2%、1.7%,测定线性可达800U/ml。标本中胆红素430μmol/L以下,血红蛋白5.5g/L以下以及甘油三酯25.0mmol/L以下对血清PON1测定均无明显影响。正常人血清PON1活性呈正态分布,参考范围为45.5~268·8U/mL。结论建立的血清PON1自动化测定方法是简单、可靠、快速、廉价的方法,适合临床实验常规测定需要。
Objective To establish and evaluate a method for the automated detection of paraoxonolipase 1 (PON1) in serum. Methods Automated determination of serum PON1 parameters was performed on Hitachi7600. The repeatability, linearity and interference analysis of this method were evaluated, and the content of PON1 in serum was analyzed. Results The total variation of PON1 was 6.5%, 4.2% and 1.7% at low, middle and high concentrations, respectively. The linearity was 800 U / ml. There was no significant difference in serum PON1 between bilirubin 430μmol / L, hemoglobin 5.5g / L and triglyceride 25.0mmol / L. Normal PON1 serum activity was normal distribution, the reference range of 45.5 ~ 268 · 8U / mL. Conclusion The automated determination of serum PON1 is a simple, reliable, rapid and inexpensive method suitable for the routine determination of clinical trials.
