目的研究低压电脉冲刺激对荷黑素瘤小鼠的抑瘤效应。方法建立BALB/c小鼠皮下B16F10黑素瘤小鼠模型,随机分为对照组与10、20、30 Hz电刺激组,电刺激组接受30 min/d、连续20 d电刺激。每日测量肿瘤体积,HE染色观察肿瘤组织学变化,分别采用实时荧光定量聚合酶链式反应检测肿瘤组织caspase-3和基质金属蛋白酶9(MMP-9)的mRNA水平,免疫组织化学法检测肿瘤组织caspase-3和MMP-9的蛋白表达。结果与对照组相比,7 d时电刺激30 Hz组肿瘤体积减小,20 d时,各电刺激组肿瘤体积减小,随时间延长抑瘤率增加;电刺激组肿瘤细胞核固缩、坏死较多;各电刺激组caspase-3 mRNA及蛋白质表达增加;MMP-9 mRNA及蛋白表达降低,caspase-3、MMP-9表达强度变化随着频率增大而增强。结论低压电刺激可上调caspase-3表达,抑制MMP-9表达,抑制肿瘤生长。
Objective To study the anti-tumor effect of low-voltage electrical pulse on mice bearing melanoma. Methods BALB / c mice model of subcutaneous B16F10 melanoma was established and randomly divided into control group and 10,20,30 Hz electrical stimulation group. The electrical stimulation group received 30 min / d electrical stimulation for 20 days. The tumor volume was measured daily, and the histological changes were observed by HE staining. The mRNA levels of caspase-3 and MMP-9 in tumor tissue were detected by real-time fluorescence quantitative polymerase chain reaction (RT-PCR) Tissue caspase-3 and MMP-9 protein expression. Results Compared with the control group, the tumor volume of 30 Hz electrical stimulation group decreased on the 7th day. At 20 days, the tumor volume of each electrical stimulation group decreased, and the tumor inhibition rate increased with time. In the electrical stimulation group, the nuclear condensation and necrosis of the tumor cell While the expression of caspase-3 mRNA and protein were increased in each group. The expression of MMP-9 mRNA and protein was decreased. The changes of caspase-3 and MMP-9 expression intensity were enhanced with increasing frequency. Conclusion Low-voltage electrical stimulation can up-regulate the expression of caspase-3, inhibit the expression of MMP-9 and inhibit tumor growth.
