目的探讨高血压对大鼠有机阴离子转运多肽(organic anion transporting polypeptides,OATPs/Oatps)及多药耐药相关蛋白(multidrug resistance-associated protein,Mrps)mRNA水平的影响。方法 14周龄的雄性自发性高血压大鼠(Spontaneous Hypertension Rat,SHR)为SHR组(SHR,n=10),同源正常的京都大鼠(Wistar-Kyoto rat,WKY)为WKY组(WKY,n=10)。常规饲养12周。每两周记录各组大鼠体重变化、Kent Scientific CODA系统检测大鼠血压变化。Real time RT-PCR检测大鼠肝脏组织中的主要摄取性转运体Oatp1a1、Oatp1a4、Oatp1a5、Oatp1b2及主要外排型转运体Mrp1、Mrp2、Mrp3、Mrp4 mRNA的水平。结果与WKY组相比,SHR组大鼠血压明显上升(P<0.05),SHR组大鼠肝脏转运体Oatp1a1、Oatp1a4、Oatp1a5、Oatp1b2、Mrp2及Mrp4 mRNA的表达量显著增高(P<0.01),而Mrp3 mRNA的表达量显著降低(P<0.01),Mrp1 mRNA的表达量无明显变化(P>0.05)。4种OATP中,Oatp1b2 mRNA在WKY(32.77%of f’-actin)及SHR(79.59%of f’-actin)中表达均最高,Oatpla5 mRNA在WKY(0.0033%of f’-actin)及SHR(0.009 2%of f’-actin)中表达均最低。4种Mrp中Mrp2 mRNA在WKY(22.46%of f’-actin)及SHR(61.04%of f’-actin)中表达均最高。结论高血压对大鼠肝脏主要摄取性转运体Oatp1a1、Oatp1a4、Oatp1a5、Oatp1b2及主要外排型转运体Mrp2、Mrp3、Mrp4 mRNA的表达有影响,而对Mrp1 mRNA的表达无影响。
Objective To investigate the effect of hypertension on mRNA levels of OATPs and Mrps in rats. Methods SHR (SHR, n = 10) and WKY (WKY) were randomly divided into WKY group (WKY group), 14 weeks old male spontaneous hypertensive rat (SHR) , n = 10). Routinely for 12 weeks. The body weight of rats in each group was recorded every two weeks. The blood pressure was measured by Kent Scientific CODA system. Real time RT-PCR was used to detect the levels of Oatp1a1, Oatp1a4, Oatp1a5, Oatp1b2 and Mrp1, Mrp2, Mrp3 and Mrp4 mRNAs in rat liver tissues. Results Compared with WKY group, the blood pressure in SHR group was significantly increased (P <0.05). The expression of Oatp1a1, Oatp1a4, Oatp1a5, Oatp1b2, Mrp2 and Mrp4 mRNA in SHR group were significantly increased (P <0.01) Mrp3 mRNA expression was significantly decreased (P <0.01), Mrp1 mRNA expression was not significantly changed (P> 0.05). Among the four OATPs, Oatp1b2 mRNA was the highest expressed in WKY (32.77% of f’-actin) and SHR (79.59% of f’-actin) and Oatpla5 mRNA was the highest in WKY (0.0033% of f’-actin) 0.009 2% of f’-actin). Mrp2 mRNA expression was the highest in WKY (22.46% of f’-actin) and SHR (61.04% of f’-actin) of the four kinds of Mrp. Conclusion Hypertension affects the expressions of Oatp1a1, Oatp1a4, Oatp1a5 and Oatp1b2 and the major efflux transporters Mrp2, Mrp3 and Mrp4 in rat liver, but has no effect on Mrp1 mRNA expression.
