目的通过建立便秘大鼠模型,检测其衰老相关指标,探讨便秘与衰老的相关性。方法在饲料中加入大黄,通过观察大鼠24 h粪便形态、重量、数量(粒数)以及测定肠道传输功能,确定泻药性便秘大鼠造模成功。比较正常组与模型组血清中超氧化物歧化酶(SOD)含量,计算胸腺、脾脏指数以及大脑海马体中klotho蛋白表达水平。结果与正常组大鼠相比,模型组大鼠24 h粪便重量、数量,肠道活性炭推进百分比,血清SOD水平,胸腺指数,大脑海马体klotho蛋白浓度、klotho目的蛋白光密度值和klothoβctin光密度值均显著降低(P<0.01或P<0.05)。结论便秘与大鼠衰老指标有相关性,其机理尚需进一步研究。
Objective To establish a rat model of constipation and test its aging-related indexes to explore the correlation between constipation and aging. Methods The rhubarb was added into the feed, and the stool form, weight, quantity (number of grains) and intestinal transit function of the rats were observed 24 h after the operation to establish the model of laxative constipation in rats. The content of superoxide dismutase (SOD) in serum of normal group and model group was compared to calculate the thymus and spleen index and klotho protein expression in hippocampus. Results Compared with the normal group, the stool weight, number, the percentage of intestinal activated carbon, the level of serum SOD, the thymus index, the concentration of klotho protein, the klotho protein and the klothoβctin optical density Values were significantly lower (P <0.01 or P <0.05). Conclusions There is a correlation between constipation and aging index in rats. The mechanism needs further study.
