目的观察粉防己碱(Tetrandrine,Tet)对大鼠碱烧伤后角膜新生血管(corneal neovascularization,CNV)生长及基质金属蛋白酶(matrix metallopretcinases-2,MMP-2)表达的影响,探讨其抗血管生成的作用机制。方法建立SD大鼠角膜碱烧伤模型。实验用药后裂隙灯下观察各组大鼠角膜新生血管的生长情况;并用免疫组织化学法测定MMP-2表达。结果正常对照组角膜透明,MMP-2分泌较少;碱烧伤对照组新生血管最多;各时间点Tet治疗组角膜新生血管发展均较碱烧伤对照组轻,MMP-2的表达亦较碱烧伤对照组显著降低。三组各时间段MMP-2的表达均有统计学意义。结论粉防己碱可能通过抑制MMP-2的表达而抑制新生血管的生成。
Objective To investigate the effect of Tetrandrine (Tet) on the growth of corneal neovascularization (CNV) and the expression of matrix metalloproteinase-2 (MMP-2) after alkali burn in rats, Mechanism. Methods The model of corneal alkali burn in SD rats was established. The rabbits were sacrificed and the corneal neovascularization was observed under the slit lamp. The expression of MMP-2 was detected by immunohistochemistry. Results The cornea of the normal control group had less corneal secretion and less secretion of MMP-2. The alkali-burn control group had the most angiogenesis. The development of corneal neovascularization in Tet-treated group was lighter than that in the alkali burn group at each time point, and the expression of MMP- Group decreased significantly. The expression of MMP-2 in three groups was statistically significant. Conclusion Tetrandrine may inhibit neovascularization by inhibiting the expression of MMP-2.
