采用稀释涂布法对芦笋老茎堆肥不同发酵阶段6个样品中的嗜热可培养放线菌进行分离,经纯化得到菌落形态有明显区别的23个菌株;根据16S rDNA序列分析结果,其中,4株的序列与白浅灰链霉菌Streptomyces albogriseolus的同源性最高,5株的序列与热普通链霉菌Streptomyces thermovulgaris的同源性最高,2株的序列与假浅灰链霉菌Streptomyces pseudogriseolus的同源性最高,1株的序列与刺棘链霉菌Streptomyces espinosus的同源性最高,有11株在GenBank数据库中未找到与其相似的已知链霉菌种的序列,分类地位待定。结果说明,芦笋老茎堆肥中优势的嗜热可培养放线菌主要是白浅灰链霉菌和热普通链霉菌。
The thermophilic actinomycetes isolated from 6 samples of different stages of the composting of Asparagus officinalis (Asparagus officinalis L.) were separated by dilution coating, and 23 strains with distinct colony morphology were obtained. According to 16S rDNA sequence analysis, 4 strains had the highest homology with Streptomyces albogriseolus, 5 strains had the highest homology with Streptomyces thermovulgaris, 2 strains had homology with Streptomyces pseudogriseolus The highest homology was found between one strain and Streptomyces espinosus, and 11 strains of Streptomyces espinosus were not found in GenBank. The results show that the dominant thermophilic cultivated actinomycetes in the compost of old stems of Asparagus officinalis are mainly Streptomyces lividans and Streptomyces vulgaris.
