The water-soluble extract of Salvia miltiorrhiza has been found to inhibit the action of cyclic adenylate phosphodiesterase (cAPD) in vitro. This inhibition was universal for the animals and tissues tested, but the cAPD was most sensitive in the rabbit house, brain, bladder, and lung. Each determination containing micrograms of water-soluble organic compounds of Salvia miltiorrhiza can produce a significant inhibitory effect, the dose increased to 1-2 mg completely inhibited. The protein in rabbit brain homogenate had protective effect on its cAPD. The study of the kinetics of enzyme action indicated that the water-soluble constituents of Salvia miltiorrhiza show a non-competitive inhibition of rabbit cAPD and are irreversibly inhibited. The water soluble extract of Salvia miltiorrhiza can be used to detect 16 spots using silica gel G thin-layer chromatography. Fraction 4 (mainly spot 13) isolated with organic solvents had the strongest inhibitory effect on cAPD in rabbit brains, with a 50% inhibition of 8 μg/ml, 32 times stronger than theophylline. Salvia miltiorrhiza water-soluble ingredients may exert some physiological effects by inhibiting cAPD.
