对T细胞发育的研究表明:胸腺是T细胞发育的主要场所,来源于骨髓的pro-T细胞进入胸腺,在胸腺微环境的作用下,经阳性和阴性选择逐渐发育为成熟的T细胞输出胸腺。但目前对T细胞在骨髓内是如何由造血干细胞分化成pro-T细胞的发育过程还不太清楚,骨髓微环境与此过程密切相关。现在的研究表明骨髓造血微环境主要是由骨髓基质细胞(Bone Marrow Stromal Cell,BMSC)及其分泌的细胞外基质、细胞因子三者共同组成,影响着造血细胞的粘附、增殖和分化。其中骨髓基质细胞是关键成分,主要包括成纤维细胞、脂肪细胞、巨噬细胞、内皮细胞和上皮细胞等。各种基质细胞网状分隔构成微环境支架,造血细胞在此微环境支架中与基质细胞相互作用并接受基质细胞分泌的造血因子的刺激而增殖分化。研究T细胞在骨髓内发育途径及分化机理,须分析BMSC类型、功能及其与发育中的T细胞的相互作用效应,其体外培养可以仔细分析骨髓微环境的作用。为此,我们继建成了小鼠胸腺基质细胞系以后,开始建立小鼠骨髓基质细胞系,本文报道所建骨髓基质细胞系BMSC1,并对其进行了细胞学鉴定分析。
T cell development studies have shown that: the thymus is the main site of T cell development, pro-T cells derived from the bone marrow into the thymus, thymus microenvironment in the positive and negative selection gradually developed into mature T cells output thymus . However, it is unclear how the T cells are differentiated into pro-T cells from the hematopoietic stem cells in the bone marrow. The bone marrow microenvironment is closely related to this process. The current research shows that the bone marrow microenvironment mainly consists of Bone Marrow Stromal Cell (BMSC) and its secreted extracellular matrix and cytokines, which affect the adhesion, proliferation and differentiation of hematopoietic cells. Bone marrow stromal cells are the key components, including fibroblasts, adipocytes, macrophages, endothelial cells and epithelial cells. A variety of stromal cells reticulate to form microenvironment scaffolds, hematopoietic cells proliferate and differentiate in this microenvironment scaffold by interacting with stromal cells and stimulated by hematopoietic factors secreted by stromal cells. To study the pathways and differentiation mechanism of T cells in bone marrow, we need to analyze the type and function of BMSC and its interaction with developing T cells. The in vitro culture can analyze the effect of bone marrow microenvironment. Therefore, we established the mouse bone marrow stromal cell line after the establishment of the mouse thymus stromal cell line. We report the construction of the bone marrow stromal cell line BMSC1 and analyze the cytology results.
