生物工程制药是70年代以后发展起来的新兴科技.是国际上公认的最有希望的医药发展方向,世界各国都投巨资以开发生产,目前已有很多产品趋向成熟并投向市场。生物工程药物以其疗效高、毒副作用相对较小而受到广大临床医生的青睐,如何正确合理使用生物工程药物也当然成为临床药师、医生需要了解、掌握的问题之一。本刊自本期起开设“生物工程制药”栏目,重点介绍生物工程制药(包括生物化学制药)的临床应用,通过栏目能使生物工程药物广泛使用于临床,造福广大患者是我们的期望所在,更欢迎广大读者赐稿,共同把栏目办好。 Bioengineering is an emerging technology developed since the 1970s, which is recognized internationally as the most promising direction for the development of medicine. All countries in the world have invested heavily in developing their products. At present, many products have matured and are ready for market. Bioengineered drugs are favored by clinicians for their high efficacy and relatively small toxic and side effects. Therefore, how to use bioengineered drugs correctly and correctly has certainly become a problem for clinical pharmacists and doctors to understand and master. 目的 构建表达外源性胶质细胞源性神经营养因子(GDNF)的转基因大鼠骨髓基质干细胞(BMSCs),拟应用于脊髓损伤的基因治疗研究.方法提取新生大鼠肾组织mRNA,用逆转录PCR法扩增GDNF全长cDNA,构建其真核表达载体PEG-GFP-GDNF,酶切及测序鉴定;然后转染原代培养的大鼠BMSCs,荧光免疫细胞化学及细胞形态学检测GDNF的表达.结果扩增到全长687 bp序列准确的GDNF编码片段,
