目的探讨米诺地尔对增生性瘢痕成纤维细胞增殖和细胞结构的影响。方法体外原代培养增生性瘢痕成纤维细胞,将0.125~4.0mmol/L的米诺地尔分别作用增生性瘢痕成纤维细胞12h,24h,48h后,采用MTT、流式细胞仪、透射电镜方法检测细胞的增殖活性及其超微结构的变化。结果MTT发现0.25~4.0mmol/L浓度的米诺地尔对增生性瘢痕成纤维细胞的增殖有抑制作用。用药48h后,米诺地尔组细胞的体积变小,核变小,核仁变小,胞内有许多溶酶体出现,未见到明显的细胞凋亡;用药48h后流式细胞仪检测发现,细胞周期中用药组比对照组S期细胞减少10.1%,G1期增加了10.4%。结论米诺地尔对增生性瘢痕成纤维细胞的增殖有抑制作用。其作用机制:米诺地尔抑制胶原的合成,另外可能增加K+外流,使细胞膜超极化,减少Ca2+内流使细胞的Ca2+浓度下降及降低细胞内的自由基生成来抑制瘢痕成纤维细胞的增殖。
Objective To investigate the effect of minoxidil on the proliferation and cell structure of hypertrophic scar fibroblasts. Methods Hypertrophic scar fibroblasts were cultured in vitro and the proliferative scar fibroblasts were treated with minoxidil 0.125 ~ 4.0mmol / L for 12h. Results MTT showed that minoxidil concentration of 0.25-4.0 mmol / L inhibited the proliferation of hypertrophic scar fibroblasts. Flow cytometry It was found that the cells in the cell cycle reduced by 10.1% in the S phase and increased in the G1 phase by 10.4% compared with the control group. Conclusion Minoxidil can inhibit the proliferation of hypertrophic scar fibroblasts. Its mechanism of action: Minoxidil inhibits the synthesis of collagen, the other may increase the K + outflow, the membrane hyperpolarization, reducing Ca2 influx of cells to reduce the concentration of Ca2 + and reduce intracellular free radical generation to inhibit scar fibroblasts proliferation.
