多抗乙素的抑瘤机制及与阿霉素、5-氟尿嘧啶联用的抑瘤效果

用患有进展期S180肉瘤的小鼠进行的实验证实,多抗乙素(PB)对小鼠S180肉瘤有明显的抑制作用,使肿瘤生长延缓,坏死显著增加,瘤周瘤内淋巴细胞、巨噬细胞浸润明显增多,荷瘤鼠脾重明显增加,脾白髓动脉周围淋巴鞘及生发中心均增大,同时表明经PB治疗的肿瘤组织显示明显的出血坏死,坏死周边部瘤组织明显充血水肿,有更多的中性粒细胞浸润及小血管内血栓形成。封闭巨噬细胞后PB的抑瘤作用消失。结合以前的研究说明:PB的抑瘤作用主要是通过活化的免疫活性细胞(巨噬细胞、T细胞等)产生的TNF等细胞活性物质实现的。PB与阿霉素或5-氟尿嘧啶联用后,在抑瘤作用、肿瘤坏死比率、瘤周瘤内免疫活性细胞浸润及脾重增加等方面明显优于其单纯应用,证明它们之间存在明显的协同作用,且PB能明显减轻阿霉素、5-氟尿嘧啶对骨髓的抑制作用。PB+ADM组与PB组或ADM组相比,P<0.01;PB+5-Fu组与PB组或5-Fu组相比,P<0.01。从表1可知,PB、ADM、5-Fu对小鼠S180肉瘤均具有显著的抑制作用,ADM或5-Fu与PB联用后,抑瘤作用明显优于ADM或5-Fu或PB单用,且PB与5-Fu联用后有2例皮下肿瘤消失。2用药第4天各组月中? Experiments with mice with advanced S180 sarcoma confirmed that polymorphonium B (PB) has a significant inhibitory effect on S180 sarcoma in mice, causing tumor growth retardation and necrosis to increase significantly. The number of phagocytic infiltrates increased significantly, and the spleen weight of tumor-bearing mice significantly increased. The lymphatic sheath and germinal centers around the white pulp of the spleen increased, indicating that the tumor tissues treated with PB showed significant hemorrhage and necrosis. There is more neutrophil infiltration and small blood vessel thrombosis. The inhibitory effect of PB on the macrophages disappeared. Combined with previous studies, it was demonstrated that the anti-tumor effect of PB is mainly achieved by active cellular active substances such as TNF produced by activated immune cells (macrophages, T cells, etc.). When PB was combined with doxorubicin or 5-fluorouracil, its antitumor effect, ratio of tumor necrosis, immunologically active cell infiltration in tumors, and increase of spleen weight were significantly better than those of pure application. It was proved that there was obvious difference between them. Synergy, and PB can significantly reduce the inhibition of adriamycin, 5-fluorouracil on bone marrow. The PB+ADM group had P<0.01 compared with the PB group or the ADM group; the PB+5-Fu group had P<0.01 compared with the PB group or the 5-Fu group. As can be seen from Table 1, PB, ADM, and 5-Fu have significant inhibitory effects on S180 sarcoma in mice. After ADM or 5-Fu is combined with PB, the anti-tumor effect is significantly better than that of ADM or 5-Fu or PB alone.
