目的:建立以HPLC-荧光检测法测定人血浆中厄贝沙坦浓度的方法。方法:色谱柱为Diamonsil C18,流动相为0.02mol方芸L-1磷酸二氢钾-乙腈(50∶50),荧光激发波长为250nm,发射波长为375nm,流速为1.0mL·min-1,柱温为40℃。结果:厄贝沙坦检测浓度在1.0~1000ng·mL-1范围内线性关系良好(r=0.9998);低、中、高3种浓度(2.0、50.0、500.00ng·mL-1)的平均相对回收率分别为97.4%、106.1%、101.6%,日内、日间RSD均小于10%。结论:本方法简便、快速、准确、特异性和灵敏度高,可用于厄贝沙坦的血药浓度测定和药动学研究。
Objective: To establish a method for the determination of irbesartan in human plasma by HPLC-fluorescence detection. METHODS: Diamonsil C18 was used as the mobile phase, and the mobile phase consisted of 0.02mol Fang Li L-1 monobasic potassium phosphate-acetonitrile (50:50). The fluorescence excitation wavelength was 250nm, the emission wavelength was 375nm and the flow rate was 1.0mL · min- The column temperature was 40 ° C. Results: The linearity of irbesartan in the range of 1.0-1000 ng · mL-1 was good (r = 0.9998). The average relative concentrations of 2.0, 5.0, 500.00 ng · mL- The recoveries were 97.4%, 106.1% and 101.6%, respectively. The daily and daily RSDs were less than 10%. Conclusion: The method is simple, rapid, accurate, specific and sensitive and can be used for determination of irbesartan and pharmacokinetics.
