目的探讨β1,3-N-乙酰氨基葡萄糖转移酶/β1,3半乳糖基转移酶(β3GnT8/β3GalT7)与肿瘤浸润、转移的相关性。方法构建并鉴定β3GnT8正义和反义表达载体。将空载体(A组)、正义(B组)及反义(C组)重组载体分别转染人胃癌SGC7901细胞中;另设SGC7901细胞为对照(D)组。采用RT-PCR和Western blot法分别检测四组细胞中基质金属蛋白酶-2(MMP-2)、MMP-14和转化生长因子β1(TGF-β1)的mRNA和蛋白表达。结果获得β3GnT8/β3GalT7基因正义和反义表达载体,并成功转染SGC7901细胞。D组TGF-β1、MMP-2和MMP-14表达低于B组,但高于C组。结论β3GnT8/β3GalT7表达与MMP-2、MMP-14和TGF-β1表达呈正相关,故可能参与肿瘤的发生、发展过程。
Objective To investigate the relationship between β1,3-N-acetylglucosaminyltransferase / β1,3-galactosyltransferase (β3GnT8 / β3GalT7) and tumor invasion and metastasis. Methods Construction and identification of β3GnT8 sense and antisense expression vector. The empty vector (group A), sense (group B) and antisense (group C) recombinant vector were transfected into human gastric cancer SGC7901 cells; SGC7901 cells as a control group (D). The mRNA and protein expressions of MMP-2, MMP-14 and TGF-β1 were detected by RT-PCR and Western blot respectively. Results The sense and antisense expression vectors of β3GnT8 / β3GalT7 gene were obtained and transfected into SGC7901 cells successfully. The expression of TGF-β1, MMP-2 and MMP-14 in group D was lower than that in group B, but higher than that in group C. Conclusion The expression of β3GnT8 / β3GalT7 is positively correlated with the expression of MMP-2, MMP-14 and TGF-β1, which may be involved in the occurrence and development of tumors.
