目的探讨人脐带间充质干细胞(h UC-MSCs)修复子痫前期重度患者血清诱导的内皮损伤模型的能力。方法将HUVECs通过transwell小室与h UC-MSCs进行共培养作用,分为对照组、实验组、干细胞干预组,观察h UC-MSCs对子痫前期重度患者血清损伤内皮细胞增殖的影响;检测各组ICAM-1及VCAM-1含量变化。结果CCK-8检测可见实验组较对照组细胞增殖明显受到抑制(P<0.05),在作用48h较作用24h抑制增殖效果明显。细胞接种24h及以上干细胞干预组较实验组内皮细胞增殖有所好转(P<0.05);ELISA检测可见实验组较对照组ICAM-1及VCAM-1均明显升高(P<0.05);经h UC-MSCs干预后,ICAM-1及VCAM-1均较实验组有所下降(P<0.05)。结论 h UC-MSCs对子痫前期重度患者血清损伤后的内皮细胞损伤模型有一定的修复作用。
Objective To investigate the ability of human umbilical cord mesenchymal stem cells (h UC-MSCs) to repair serum-induced endothelial injury in severe preeclampsia patients. Methods HUVECs were co-cultured with h UC-MSCs via transwell chamber and divided into control group, experimental group and stem cell intervention group. The effects of h UC-MSCs on the proliferation of endothelial cells in severe preeclampsia patients were observed. ICAM-1 and VCAM-1 content changes. Results The results of CCK-8 showed that the proliferation of cells in experimental group was significantly inhibited (P <0.05), and the effect of inhibiting proliferation was obvious at 48h after treated with CCK-8. The number of ICAM-1 and VCAM-1 in the experimental group was significantly higher than that in the control group (P <0.05). After the cells were inoculated for 24h and above, After UC-MSCs intervention, ICAM-1 and VCAM-1 decreased compared with the experimental group (P <0.05). Conclusion h UC-MSCs can repair endothelial cell injury model in severe preeclampsia patients after serum-induced injury.
