目的:建立贯叶金丝桃中芦丁与金丝桃苷的含量测定方法。方法:采用高效液相色谱仪,使用Diamonsil C18柱(250mm×4.6 mm,5μm),流动相为甲醇-乙腈-四氢呋喃-0.4%醋酸(1∶1∶18∶80),流速为0.7 ml·min-1,检测波长为358 nm,柱温为30℃。结果:芦丁在0.206~1.236μg范围内有良好的线性关系,r=0.999 9。金丝桃苷在0.144~0.864μg范围内有良好的线性关系,r=0.999 8;芦丁的平均加样回收率为98.57%,RSD为0.94%,金丝桃苷的平均加样回收率为98.57%,RSD为0.94%。结论:该方法简便快捷,准确度高。
Objective: To establish a method for the determination of rutin and hyperoside in Hypericum perforatum. Methods: Diamonsil C18 column (250 mm × 4.6 mm, 5 μm) was used with a mobile phase of methanol - acetonitrile - tetrahydrofuran - 0.4% acetic acid (1:1:18:80) at a flow rate of 0.7 ml · min -1, the detection wavelength is 358 nm, the column temperature is 30 ℃. Results: There was a good linear relationship between rutin and 0.206 ~ 1.236μg, r = 0.999 9. Hyperoside had a good linear relationship in the range of 0.144 ~ 0.864 μg, r = 0.999 8. The average recoveries of rutin were 98.57% with RSD of 0.94%. The average recoveries of hyperoside were 98.57%, RSD 0.94%. Conclusion: This method is simple and quick with high accuracy.
