目的研究Tiam1-SiRNA质粒转染HCT116细胞对裸鼠移植瘤中淋巴管生成的影响。探讨Tiam1与结肠癌淋巴管生成的关系。方法裸鼠10只随机分为对照组(n=5)和Tiam1-SiRNA组(n=5),将含Tiam1-SiRNA质粒的HCT116细胞进行裸鼠皮下接种,5周后处死动物,留取肿瘤标本。利用Western blot和免疫组化SP法检测Tiam1、Rac1、VEGF-C/D蛋白水平的表达,并进行肿瘤淋巴管染色计数。结果干扰组移植瘤中Tiam1、Rac1、VEGF-C、VEGF-D蛋白表达较对照组显著降低(P<0.01),肿瘤内淋巴管生成也显著减少(P<0.01)。结论通过RNA干扰技术抑制Tiam1的表达,在裸鼠体内可显著抑制移植瘤组织内淋巴管生成。
Objective To investigate the effect of Tiam1-SiRNA plasmid transfected HCT116 cells on lymphangiogenesis in nude mice xenografts. To investigate the relationship between Tiam1 and lymphangiogenesis in colon cancer. Methods Ten nude mice were randomly divided into control group (n = 5) and Tiam1-SiRNA group (n = 5). HCT116 cells containing Tiam1-SiRNA plasmid were inoculated subcutaneously in nude mice. specimen. The expression of Tiam1, Rac1, VEGF-C / D protein was detected by Western blot and immunohistochemistry SP method, and lymphatic vessel staining was counted. Results The expression of Tiam1, Rac1, VEGF-C and VEGF-D in the transplanted tumor was significantly lower than that in the control group (P <0.01), and the lymphangiogenesis was also significantly reduced in the transplanted tumor (P <0.01). Conclusion Inhibition of Tiam1 expression by RNA interference can significantly inhibit the lymphangiogenesis in xenografts in nude mice.
