目的利用AFLP技术分析香鳞毛蕨种质资源多样性。方法对46个香鳞毛蕨个体进行AFLP分析,利用NTSYS-PC 2.10、PopGene32软件分析各居群的遗传多样性。结果香鳞毛蕨居群多态位点百分率(PPB)为76.67%;观测等位基因数(Na)为1.766 7,有效等位基因数(Ne)为1.504 6,Ne′is基因多样性指数(H)为0.292 1,Shannon信息指数(I)为0.431 6;遗传分化指数(Gst)为0.412 5。结论香鳞毛蕨遗传多样性较高。群体内的基因多样性(Hs)为0.170 5,群体间的基因多样性(Dst)为0.119 8,且由于生境特殊性,对香鳞毛蕨资源应采取就地保护策略。
OBJECTIVE: To analyze the diversity of Fraxinus mandshurica resources by using AFLP technique. Methods AFLP analysis was performed on 46 individuals of Fraxinus mandshurica and the genetic diversity of each population was analyzed by NTSYS-PC 2.10 and PopGene32 software. Results The percentage of polymorphic loci (PPB) was 76.67%, the number of observed alleles (Na) was 1.766 7, the number of effective alleles (Ne) was 1.504 6, the index of Ne’is gene diversity (H) was 0.292 1, Shannon’s information index (I) was 0.431 6, and the genetic differentiation index (Gst) was 0.412 5. Conclusion There is a high genetic diversity in Fraxinus officinalis. The population genetic diversity (Hs) was 0.170 5, and the genetic diversity among populations (Dst) was 0.119 8. Due to the particularity of habitat, the strategy of in situ conservation should be taken for the resources of A. fragrans.
