目的:检测胃癌组织标本β-catenin mRNA表达水平,并探讨其临床意义。方法:以β-actin为内参照,采用实时荧光定量PCR技术检测45例胃癌患者术后胃癌组织标本、肿瘤边缘5 cm外的正常组织β-catenin基因mRNA的表达水平,并进行比较。结果:胃癌患者术后胃癌组织标本β-catenin mRNA表达水平较正常组织标本高(P<0.01);肠型胃癌β-catenin mRNA表达水平高于弥漫型胃癌(P<0.05);TNM分期早期胃癌β-catenin mRNA表达水平低于晚期(P<0.05);淋巴结有转移者高于无转移者(P<0.01),且与浸润深度相关(P<0.01);胃癌β-catenin mRNA表达水平与患者性别、年龄、肿瘤大小及分化程度无关。结论:β-catenin mRNA在胃癌组织中呈高表达,是评价疾病发生、发展的良好指标。
Objective: To detect the expression of β-catenin mRNA in gastric cancer tissue and to explore its clinical significance. Methods: The expression of β-catenin mRNA in 45 specimens of gastric cancer tissues and 5-cm margin of tumor was detected by real-time fluorescence quantitative PCR with β-actin as internal reference. Results: The expression of β-catenin mRNA in gastric cancer tissues was significantly higher than that in normal tissues (P <0.01). The expression of β-catenin mRNA in gastric cancer was higher than that in diffuse gastric cancer (P <0.05) The expression of β-catenin mRNA in gastric cancer was significantly lower than that in advanced stage (P <0.05). The level of β-catenin mRNA in gastric cancer was significantly higher than that in patients without lymph node metastasis (P <0.01) Sex, age, tumor size and degree of differentiation have nothing to do. Conclusion: β-catenin mRNA is highly expressed in gastric cancer and is a good indicator to evaluate the occurrence and development of the disease.
