在改良MS培养基上 ,全黑暗条件下培养未经任何处理的玉米种子萌发的茎尖 ,可诱导雌雄花序的发生 .培养基 6 BA浓度 2 0mg/L时 ,自交系 5 15 ,8112及其杂交后代的茎尖分生组织经过 5~ 6周的培养 ,可直接分化产生雌雄花序 ;培养基的激素组合为 6 BA 1.0mg/L +2 ,4 D 0 .2mg/L ,供试的 6个自交系的茎尖膨大生长 ,产生较多的丛生芽 ,在转入添加 6 BA 1.0mg/L +IBA 0 .2mg/L的培养基上后 ,培养物在继续产生丛生芽的同时 ,分化出大量的雌雄花序 .雌、雄花序发生过程与田间植株的基本相同 ,但雌穗远多于雄穗 .适宜外源激素是诱导雌、雄花序发生所必需的 ,基因型决定了外植体对诱导条件的反应 .由于离体茎尖能直接产生生殖器官 ,推测玉米营养生长和生殖器官发生被不同调控机制所控制 .
