目的探讨缺氧诱导因子1α(HIF-1α)对宫颈癌细胞的生物学行为的影响。方法选用Hela细胞、空载体EGFP-C1转染Hela、EGFP-HIF-1α反义Hela细胞,在37℃,5%CO2,1%O2培养环境中培养细胞对照;通过Western blotting检测各组细胞内HIF-1α蛋白表达,用流式细胞仪检测各组细胞的凋亡率、软琼脂克隆形成方法观察各组细胞的增殖、Transwell侵袭小室方法观察各组细胞的侵袭能力的改变。结果蛋白免疫印迹结果显示,在低氧环境中Hela细胞和仅转染EGFP-C1质粒的Hela细胞中有HIF-1α蛋白的表达条带,而EGFP-HIF-1α反义Hela细胞无表达。并且在低氧环境中EGFP-HIF-1α反义Hela细胞与Hela细胞和仅转染EGFP-C1质粒的Hela细胞相比,其细胞的凋亡率增加、细胞的侵袭能力及细胞集落的形成被抑制,差异均有统计学意义(P<0.01)。结论 HIF-1α能够对宫颈癌Hela细胞的恶性生物学行为产生影响,包括抗凋亡、促增殖、侵袭转移等,且在体外抑制HIF-1α的表达对宫颈癌细胞的凋亡有抑制作用。
Objective To investigate the effect of hypoxia inducible factor 1α (HIF-1α) on the biological behavior of cervical cancer cells. Methods Hela cells and empty vector EGFP-C1 were used to transfect Hela and EGFP-HIF-1α antisense Hela cells. The cells were cultured in 37 ℃, 5% CO2, 1% O2 culture environment. HIF-1αprotein expression, the apoptosis rate of each group of cells was detected by flow cytometry, the proliferation of each group of cells was observed by soft agar colony formation, and the invasion ability of cells in each group was observed by Transwell invasion chamber method. Results Western blotting showed that HIF-1α protein was expressed in Hela cells and Hela cells transfected with EGFP-C1 plasmid only in hypoxic environment, while EGFP-HIF-1α antisense Hela cells showed no expression. And in hypoxia environment, the apoptosis rate of EGFP-HIF-1α antisense Hela cells increased compared with Hela cells and Hela cells transfected with EGFP-C1 plasmids only. The invasive ability of cells and the formation of colony Inhibition, differences were statistically significant (P <0.01). Conclusion HIF-1α can affect the malignant biological behavior of cervical cancer Hela cells, including anti-apoptosis, proliferation, invasion and metastasis, and inhibit the expression of HIF-1α on cervical cancer cells in vitro.
