目的 了解人类良、恶性乳腺病变组织中生长抑素受体亚型 2 (SSTR2 )mRNA的表达情况 ,并探讨其与雌激素受体(ER)、孕激素受体 (PR)的相关性。方法 收集 2 3例乳腺癌、16例乳腺增生病和 9例乳腺纤维腺瘤手术标本 ,采用多相寡核苷酸探针原位杂交法检测SSTR2mRNA表达 ,免疫组化法检测ER、PR状况 ,并用图像分析仪进行SSTR2mRNA相对含量测定。结果 SSTR2mRNA在乳腺癌表达的阳性率 (87% )和相对含量 (0 .4 7)均明显高于乳腺良性病变 (6 4 % ,0 .2 6 ) (P <0 .0 5 )。乳腺良性病变中SSTR2mRNA表达与ER呈正相关 (P <0 .0 5 ) ;在乳腺癌中SSTR2mRNA表达与ER、PR均呈正相关(P <0 .0 5 )。结论 SSTR2mRNA在乳腺良、恶性病变组织中普遍表达 ,恶性高于良性 ;SSTR2mRNA表达与ER、PR具有相关性 ,提示对ER阳性乳腺癌采用抗雌激素与SST类似物联合治疗的可行性。
Objective To investigate the expression of somatostatin receptor subtype 2 (SSTR2) mRNA in human benign and malignant breast lesions and to explore its correlation with estrogen receptor (ER) and progesterone receptor (PR). Methods Surgical specimens of 23 cases of breast cancer, 16 cases of breast hyperplasia and 9 cases of breast fibroadenoma were collected. The expression of SSTR2 mRNA was detected by in situ hybridization with polyphasic oligonucleotide probe. The status of ER and PR was detected by immunohistochemistry. The relative content of SSTR2 mRNA was measured using an image analyzer. Results The positive rates of SSTR2 mRNA expression in breast cancer (87%) and relative content (0.47) were significantly higher than those in benign breast lesions (64%, 0.26%) (P < 0.05). The expression of SSTR2 mRNA was positively correlated with ER in benign breast lesions (P < 0.05). The expression of SSTR2 mRNA was positively correlated with ER and PR in breast cancer (P <0. 05). Conclusion The expression of SSTR2 mRNA is common in benign and malignant lesions of breast, and the malignancy is higher than benign. The expression of SSTR2 mRNA is correlated with ER and PR, which suggests the feasibility of combining anti-estrogen and SST analogues for ER-positive breast cancer.
