考察了奥美拉唑对映体在多糖类手性色谱柱(Chiralpak AD柱、Chiralcel OJ柱、Chiralpak AS柱和Chiralcel OD柱)上的分离行为及流动相组成对对映体分离的影响,建立了血浆中奥美拉唑对映体的分析方法。血浆样品采用Agilent BON DODS-C18固相萃取柱提取,以Chiralpak AD(250mm×4.6mm,10μm)为色谱柱;流动相:V(正己烷):V(乙醇)=30:70;流速:0.5mL/min;检测波长:302nm;柱温:25℃。方法的相对回收率为(-)-(S)-奥美拉唑96.7%~100.4%,(+)-(R)-奥美拉唑97.4%~100.2%;日内RSD<4.7%,日间RSD<3.3%;奥美拉唑对映体在5~1000ng/mL范围内线性关系良好(n=5,r=0.9997),最低检测浓度(S/N=3)2ng/mL。方法可用于奥美拉唑对映体人体药代动力学研究。
The enantioseparation of enantiomers of omeprazole enantiomers on polysaccharide chiral column (Chiralpak AD column, Chiralcel OJ column, Chiralpak AS column and Chiralcel OD column) was investigated. A method for the analysis of enantiomers of omeprazole in plasma was established. The plasma samples were extracted with an Agilent BOD DODS-C18 solid-phase extraction cartridge and chromatographed on a Chiralpak AD (250 mm × 4.6 mm, 10 μm); mobile phase: V (n-hexane): V mL / min; detection wavelength: 302nm; column temperature: 25 ℃. The relative recoveries were 96.7% -100.4% for (-) - (S) -omeprazole and 97.4% -100.2% for (+) - RSD <3.3%. The enantiomers of omeprazole had a good linear relationship (n = 5, r = 0.9997) and the lowest detection concentration (Sng / n = 3) was 2 ng / mL in the range of 5 ~ 1000 ng / mL. The method can be used to study the pharmacokinetics of omeprazole enantiomers.
