采用无血清甲基纤维素半固体培养方法,对白血病细胞系和原代白血病细胞进行一次集落培养,将形成的集落细胞重新悬浮,进行二次集落培养、计数.结果白血病细胞系的二次植入效率为31.30%,原代髓系白血病细胞的二次植入效率为22.66%.提示白血病细胞存在具有高增殖能力的干细胞性细胞群;原代白血病细胞的再增殖能力低于白血病细胞系.
A serum-free methylcellulose-based semi-solid culture method was used to perform a colony culture on leukemia cell lines and primary leukemia cells, and the resulting colony cells were resuspended for secondary colony culture and counting. Results Secondary Leukemia Cell Lines The efficiency of entry was 31.30%, and the secondary implantation efficiency of primary myeloid leukemia cells was 22.66%. This suggests that there are stem cell populations with high proliferation ability in leukemic cells; the reproliferation capacity of primary leukemia cells is lower than that of leukemic cell lines.
