目的:体外建立人肝癌细胞(HepG2)胰岛素抵抗模型,并初步研究中药复方芪参龟散对胰岛素抵抗的改善作用。方法:HepG2模型建立后,应用不同浓度的芪参龟散及不同浓度的罗格列酮分别作用于胰岛素抵抗细胞24h后,运用葡萄糖氧化酶法分别监测不同浓度的上述药物成分对HepG2细胞胰岛素抵抗模型葡萄糖消耗的影响,运用MTT法对各组细胞活性进行评价。结果:人肝癌细胞(HepG2)在10-6mol/L浓度的胰岛素作用24h后,葡萄糖消耗量明显减少(P<0.01),此结果表明实验成功地诱导出稳定人肝癌细胞胰岛素抵抗模型。10-5mol/L浓度的胰岛素组表现出了更为明显的IR(P<0.01)。但是4个时间点的死亡细胞逐渐增多,成活率降低(P<0.05)。罗格列酮高、中剂量组及芪参龟散低剂量组均有改善细胞胰岛素抵抗的作用,其中1.0mg/m L浓度的罗格列酮及0.0156mg/m L浓度的芪参龟散对改善细胞IR效果较好(P<0.05)。结论:芪参龟散有改善胰岛素抵抗的作用。
OBJECTIVE: To establish a model of human hepatocellular carcinoma (HepG2) insulin resistance in vitro and to study the effect of traditional Chinese medicine Qiguan Turtle Powder on improving insulin resistance. Methods: After HepG2 model was established, different concentrations of Qiguan turtle decoction and different concentrations of rosiglitazone were respectively administered to insulin resistance cells for 24 h. Glucose oxidase method was used to monitor the insulin resistance of HepG2 cells at different concentrations Model of glucose consumption, using MTT method to evaluate the activity of each group of cells. Results: After treated with 10-6mol / L insulin for 24 h, the glucose consumption of HepG2 cells was significantly decreased (P <0.01). The results showed that the experiment successfully induced insulin resistance in human hepatocellular carcinoma cells. The 10-5 mol / L insulin group showed a more pronounced IR (P <0.01). However, the number of dead cells gradually increased and the survival rate decreased at 4 time points (P <0.05). Rosiglitazone high dose group and Qiganshuitai low dose group have the effect of improving cell insulin resistance, in which 1.0mg / m L concentration of rosiglitazone and 0.0156mg / m L concentration of Qishen turtle bulk To improve the cell IR better (P <0.05). Conclusion: Qishen turtle decoction can improve insulin resistance.
