[目的]确定黄芩总黄酮与黄芩苷提取及纯化的最佳工艺条件。[方法]以黄芩总黄酮与黄芩苷得率作为评价指标,采用单因素及正交试验进行优选。[结果]最优工艺为:取黄芩粗粉,加10倍量水,煎煮2次,1.0 h/次;离心所得上清液调pH值至1.5,60℃保温30 min,所得沉淀加适量水搅匀,调pH值至中性,加等量乙醇醇沉;上清液再调至pH值为1.5,60℃保温30 min,静置、离心所得沉淀,水洗至中性,60℃真空干燥得黄芩提取物。其中,黄芩总黄酮、黄芩苷含量分别可达94.15%、80.15%。[结论]该工艺合理、可行,适合黄芩总黄酮、黄芩苷的工业化生产。
[Objective] The research aimed to determine the optimum conditions for the extraction and purification of total flavonoids from Scutellaria baicalensis and baicalin. [Method] The yield of total flavonoids and baicalin in Scutellaria baicalensis Georgi was used as evaluation index, and single factor and orthogonal experiment were used to optimize. [Result] The optimum process was as follows: Scutellaria baicalensis Georgi, adding 10 times the amount of water, boiling twice, 1.0 h / time; the supernatant was centrifuged to adjust the pH to 1.5, 60 ℃ incubated 30 min, the resulting precipitate plus the appropriate amount Water and stir well, adjust pH to neutral, add equal volume alcohol ethanol precipitation; the supernatant was adjusted to pH 1.5, 60 ℃ incubated 30 min, standing, centrifugation of the precipitate, washed with water to neutral, 60 ℃ vacuum Dry skullcap extract. Among them, the contents of total flavonoids and baicalin in Scutellaria baicalensis were 94.15% and 80.15% respectively. [Conclusion] The process is reasonable and feasible, suitable for the industrial production of total flavonoids and baicalin of Scutellaria baicalensis Georgi.
