目的探讨新发现的肿瘤转移抑制基因人源性长寿保障基因Ⅱ型(homo sapiens longevity assurance homologue2,LASS2)与人膀胱癌细胞侵袭潜能的关系。方法通过细胞生长曲线和体外侵袭实验,确定膀胱癌BIU-87、EJ、T24和EJ-M3细胞的增殖和侵袭能力;运用实时定量PCR法和Western blot法检测4株细胞中LASS2基因的表达。结果 4株膀胱癌细胞中,EJ-M3的增殖和侵袭能力最强;增殖和侵袭能力高的细胞系EJ-M3和BIU-87的LASS2mRNA表达降低(P<0.001),而在低转移能力的人膀胱癌细胞株T24和EJ细胞中则相对较高表达。结论 LASS2基因表达的降低,可能与膀胱癌细胞的增殖和侵袭有关。
Objective To explore the relationship between the newly discovered tumor suppressor gene homo sapiens longevity assurance homologue 2 (LASS2) and invasion potential of human bladder cancer cells. Methods The proliferation and invasion ability of bladder cancer cells BIU-87, EJ, T24 and EJ-M3 were determined by cell growth curve and in vitro invasion assay. The expression of LASS2 gene in 4 cells was detected by real-time quantitative PCR and Western blot. Results Among the four bladder cancer cells, EJ-M3 had the strongest proliferation and invasion ability, while LASS2 mRNA expression was decreased in EJ-M3 and BIU-87 cell lines with high proliferative and invasive ability (P <0.001) Human bladder cancer cell lines T24 and EJ cells were relatively highly expressed. Conclusion The decrease of LASS2 gene expression may be related to the proliferation and invasion of bladder cancer cells.
