目的:研究积雪草总苷对大鼠下丘脑-垂体-肾上腺皮质轴和下丘脑-垂体-甲状腺轴功能的影响。方法:采用长期未预知应激刺激致大鼠抑郁症模型实验,积雪草总苷灌胃给药(30、60和120 mg/kg)21 d后,用放射免疫法测定大鼠血浆ACTH和血清TSH、T3、T4和rT3的浓度。结果:与正常大鼠比较,抑郁症模型组动物血浆ACTH、血清TSH、T4和rT3浓度显著降低,血清T3浓度显著升高;积雪草总苷各剂量组血浆ACTH水平、血清TSH、T4和rT3水平不同程度增加,血清T3水平减少。结论:积雪草总苷具有调节抑郁症大鼠下丘脑-垂体-肾上腺皮质轴和下丘脑-垂体-甲状腺轴功能的作用。
OBJECTIVE: To investigate the effects of Asiaticoside on the hypothalamic-pituitary-adrenocortical axis and hypothalamic-pituitary-thyroid axis in rats. METHODS: Rat model of depression induced by long-term unpredicted stress was tested. After the administration of total glycosides of asiaticoside (30, 60 and 120 mg/kg) for 21 days, the plasma ACTH levels in rats were measured by radioimmunoassay. Concentrations of serum TSH, T3, T4, and rT3. RESULTS: Compared with normal rats, the levels of plasma ACTH, serum TSH, T4, and rT3 were significantly decreased in the depressive model group, and the serum T3 concentration was significantly increased; plasma ACTH levels, serum TSH, and T4 levels in each dose group of total glycosides of Asiaticoside Different levels of rT3 increased, serum T3 levels decreased. CONCLUSION: Total glycosides of asiaticoside may play a role in regulating the functions of hypothalamic-pituitary-adrenocortical axis and hypothalamic-pituitary-thyroid axis in depressed rats.
