目的探讨羌活醇提取物对肝缺血再灌注损伤(HIRI)大鼠模型抗氧化作用的影响及机制。方法将100只大鼠随机分为空白组、模型组、丹参预处理组(DS组)和羌活预处理组(QH组)4组,后3组组内再分为缺血45min(I45)和缺血45min再灌注30min(I45R30)、缺血45min再灌注60min(I45R60)3个时间点,DS组、QH组分别给予丹参醇提取物、羌活醇提取物灌胃,均1ml/d,连续7d。行HIRI造模后分别测定各组大鼠各时点即刻血清丙氨酸氨基转移酶(ALT)水平和肝组织丙二醛(MDA)含量和谷胱甘肽过氧化物酶(GSH-Px)活力水平。结果模型组、QH组和DS组内大鼠模型I45R30、I45R60时点的血清ALT水平、MDA含量、GSH-Px活力均较I45时点明显升高(P<0.01);DS组I45R60时点大鼠肝组织中MDA含量较QH组明显降低(P<0.05);DS组I45R60时点大鼠肝组织GSH-Px活力均较模型组和QH组低(P<0.05)。结论羌活醇提取物可能是通过提高HIRI模型大鼠肝组织GSH-Px活力,增强对氧自由基的清除能力,减轻脂质过氧化损伤,从而起到保护肝脏的作用。
Objective To investigate the effects and mechanisms of Notopterygii alcohol extract on antioxidative activity in rat model of hepatic ischemia-reperfusion injury (HIRI). Methods 100 rats were randomly divided into 4 groups: blank group, model group, Danshen pretreatment group (DS group) and Notopterygium pretreatment group (QH group) The rats in DS group and QH group were dosed with Salviae miltiorrhizae alcohol extract and Notopterygii alcohol extract respectively by gavage for 1 min every 30 min (I45R30) and 60 min (I45R60) . After the HIRI model was established, the levels of serum alanine aminotransferase (ALT), malondialdehyde (MDA) and glutathione peroxidase (GSH-Px) Vitality level. Results The levels of serum ALT, MDA and GSH-Px in I45R30 and I45R60 rats in model group, QH group and DS group were significantly higher than those in I45 (P <0.01) Compared with QH group, the content of MDA in rat liver tissue was significantly lower than that in QH group (P <0.05). The activity of GSH-Px in liver tissue of I45R60 group was lower than that of model group and QH group (P <0.05). Conclusion Extract of Notopterygii alcoholic extract may play an important role in protecting the liver by increasing the activity of GSH-Px in the liver of HIRI model rats, increasing the scavenging capacity of oxygen free radicals and reducing lipid peroxidation.
