目的探讨复方番石榴制剂对2型糖尿病胰岛素抵抗大鼠空腹血糖和胰岛素敏感性的影响。方法采取高热量饲料喂养8周后腹腔注射小剂量链脲佐菌素(STZ),复制2型糖尿病(T2DM)动物模型。随机分为模型组、二甲双胍组、复方番石榴制剂高剂量组、复方番石榴制剂低剂量组。另设正常组,饲以普通饲料。造模成功后每周称量体重一次,给药6周后处死大鼠,末次给药后禁食12 h,检测空腹血糖(FBG)、血清胰岛素含量(FINS)和胰岛素敏感指数(ISI)。结果复方番石榴制剂药物治疗期间,各给药治疗组其体重均较模型组有不同程度的增长,且二甲双胍治疗组在给药治疗后的第2周开始就对体重的增长有明显的改善作用(P均<0.05)。各给药治疗组可不同程度地降低FPG、FINS,同时提高ISI(P均<0.05),其中,复方番石榴制剂高剂量组以及二甲双胍组有明显改善FPG、FINS,提高ISI的作用。结论复方番石榴制剂可改善2型糖尿病胰岛素抵抗大鼠高血糖、高胰岛素血症,提高胰岛素敏感性的作用。
Objective To investigate the effect of compound guava preparations on fasting blood glucose and insulin sensitivity in type 2 diabetic rats with insulin resistance. Methods Twenty-eight weeks after high-calorie diet feeding, low-dose streptozotocin (STZ) was injected intraperitoneally to establish a model of type 2 diabetes mellitus (T2DM). Randomly divided into model group, metformin group, high-dose guanacide compound group, low-dose guanacide compound group. Another set of normal group, fed with ordinary feed. Rats were weighed once a week after successful administration. After 6 weeks of administration, rats were killed and fasted for 12 hours after the last administration. Fasting blood glucose (FBG), insulin (FINS) and insulin sensitivity index (ISI) were measured. Results During the treatment of compound guava preparations, the body weight of each treatment group increased to some extent compared with the model group, and metformin treatment group had a significant improvement in body weight gain from the second week after treatment (P <0.05). FPG, FINS and ISI were decreased in all treatment groups (P <0.05). The high dose guanidinium compound group and metformin group significantly improved the FPG, FINS and ISI. Conclusion Compound guava preparation can improve hyperglycemia, hyperinsulinemia and insulin sensitivity in type 2 diabetic rats with insulin resistance.
