目的:研究藏药总状绿绒蒿醇提物对人白血病K562细胞增殖的影响,并初步探讨其机制。方法:MTT法检测总状绿绒蒿醇提物对K562细胞增殖的抑制作用;Hoechest 33342荧光染色观察细胞核形态学改变;流式细胞术检测凋亡率和线粒体膜电位变化。结果:一定浓度范围内的总状绿绒蒿醇提物对K562细胞的增殖有明显抑制作用(P<0.05),并呈时间及剂量依赖性;Hoechest 33342荧光染色及凋亡率检测结果显示总状绿绒蒿醇提物处理后诱导K562细胞发生凋亡;总状绿绒蒿醇提物能明显降低K562细胞线粒体膜电位。结论:总状绿绒蒿醇提物对人白血病K562细胞有增殖抑制作用,通过改变线粒体膜电位诱导肿瘤细胞凋亡。
OBJECTIVE: To study the effects of the ethanol extract of Tibetan clover on the proliferation of human leukemia K562 cells and to explore its mechanism. Methods: MTT assay was used to detect the inhibition of K562 cell proliferation by alcohol extract of Meconopsis pumila. Koehnia stomatitis staining was used to observe the changes of nuclear morphology. The apoptosis rate and mitochondrial membrane potential were detected by flow cytometry. Results: The alcohol extract of Melastoma Gentiana decreased significantly in a concentration-dependent manner (P <0.05), and showed a time-and dose-dependent manner. The results of Hoechest 33342 staining and apoptosis showed that the total The apoptosis of K562 cells was induced by ethanol extract of Meconopsis pumila and the extract of Meconopsis pumila decreased the mitochondrial membrane potential of K562 cells. CONCLUSION: Alcohol extract of Meconopsis pumila is able to inhibit proliferation of human leukemia K562 cells and induce apoptosis of tumor cells by changing mitochondrial membrane potential.
