目的通过质量考察和处方筛选,制备蓝色纳米级染色剂,使之更容易透过血脑屏障,被肿瘤细胞吸收。方法以磷脂、双氨基PEG、京尼平与胎牛血清为原料,以超声均化法制备成蓝色纳米级染色剂冻干粉,以形态、粒径为考察目标筛选制备处方。结果制备神经胶质瘤的蓝色纳米级染色剂最佳处方为大豆卵磷脂60 mg,胎牛血清900μL,双氨基PEG 10 mg·m L~(-1),京尼平1.5 mg·m L~(-1);平均粒径为71.57 nm,平均电位为49.6 m V,电镜下形态良好。结论制备的蓝色纳米级染色剂冻干粉粒径、形态符合质量要求,纳米蛋白能有效裸眼示踪脑胶质瘤。工艺方法简单、实用。
OBJECTIVE To prepare a blue nanoscale stain through quality inspection and prescribing screening to make it easier to penetrate the blood-brain barrier and be absorbed by tumor cells. Methods The phospholipids, diamino PEG, genipin and fetal bovine serum were used as raw materials to prepare the blue nanometer-scale lyophilized powder by ultrasonic homogenization. The morphology and particle size were selected as the screening targets. Results The best prescriptions for the preparation of glioma blue nanoscale stains were soy lecithin 60 mg, fetal calf serum 900 μL, bis-amino-PEG 10 mg · m L -1, and genipin 1.5 mg · m L ~ (-1). The average particle size was 71.57 nm and the average potential was 49.6 mV. The morphology was good under electron microscope. Conclusion The preparation of blue nano-scale freeze-dried powder particle size, morphology in line with quality requirements, nano-protein can effectively naked eye traced glioma. The process is simple and practical.
