目的:研究益气解毒活络方对链尿佐菌素(STZ)诱导的糖尿病肾病(DN)大鼠肾脏结缔组织生长因子(CTGF)及Ⅳ型胶原(ColⅣ)mRNA的影响。方法:Wistar大鼠90只,二次随机分组:正常组10只、模型组16只、西药组14只、预防组14只、高量组14只、低量组14只。实验结束后,检测空腹血糖(FBG)、糖化血红蛋白(HbA1c)、尿微量白蛋白(mAlb)、β2微球蛋白(β2-MG)、肌酐(Scr)、尿素氮(BUN);HE及PAS染色观察病理学形态改变;免疫组化法观察肾组织中Bcl-2、Bax蛋白表达;实时荧光定量Q-PCR法检测肾组织中CTGF及ColⅣmRNA表达。结果:模型组FBG、HbA1c、mAlb、β2-MG、BUN明显升高(P<0.01),Scr无明显变化,肾脏病理改变明显,Bcl-2表达下降、Bax表达增加,CTGF、ColⅣmRNA含量明显上升(P<0.01),各治疗组上述指标明显改善。结论:益气解毒活络方可能通过抑制Bax表达,提高Bcl-2表达,降低mAlb,减少肾组织CTGF及ColⅣmRNA表达,从而保护肾脏。
Objective: To study the effects of Yiqi Jiedu Huoxue Huoxue Recipe on the expression of connective tissue growth factor (CTGF) and collagen Ⅳ (Col Ⅳ) in rats with streptozotocin (STZ) induced diabetic nephropathy (DN). Methods: Ninety Wistar rats were randomly divided into two groups: normal group (n = 10), model group (n = 16), western medicine group (n = 14), prevention group (n = 14), high dose group (n = 14) and low dose group (n = 14). After the experiment, FBG, HbA1c, mAlb, β2-MG, Scr and BUN were detected; HE and PAS staining The pathological morphological changes were observed. The expression of Bcl-2 and Bax protein in renal tissues was detected by immunohistochemistry. The expression of CTGF and ColⅣ mRNA in renal tissues was detected by real-time fluorescence quantitative Q-PCR. Results: The levels of FBG, HbA1c, mAlb, β2-MG and BUN in the model group were significantly increased (P <0.01), while there was no significant change in Scr. The pathological changes of kidney were obvious, the expressions of Bcl-2, Bax and CTGF, ColⅣ mRNA were significantly increased (P <0.01), the treatment group significantly improved the above indicators. Conclusion: Yiqi Jiedu Huoxue Prescription can protect the kidneys by inhibiting Bax expression, increasing Bcl-2 expression, decreasing mAlb, decreasing CTGF and Col Ⅳ mRNA expression in renal tissues.
