为了研究粒细胞集落刺激因子(G-CSF)动员对供者T淋巴细胞亚群功能和急性移植物抗宿主病(aGVHD)的影响,对20例供者G-CSF动员前后的外周血样品,加入佛波醇酯、伊能霉素、莫能霉素体外刺激培养4小时,以3色荧光标记法流式细胞术检测T细胞亚群Ⅰ(分泌INF-γ、INF-α)、Ⅱ型(分泌IL-4、IL-5和IL-10)细胞比例。结果表明:G-CSF动员前外周血CD3+IFN-γ+、CD4+IFN-γ+和CD8+IFN-γ+T细胞的比例分别为3.2%(0%-45.9%)、1.3%(0%-23.8%)和1.5%(0%-22.2%),G-CSF动员后前述细胞比例显著升高,分别为19.2%(0%-53.9%)、9.5%(0%-49.5%)和7.5%(0%-38.1%)。动员前CD3+IL-2+、CD4+IL-2+、CD8+IL-2+T细胞比例分别为1.5%(0%-31%)、0.8%(0%-30.0%)和0%(0%-5.3%),动员后比例亦明显升高,分别为25.7%(0%-51%)、19.8%(0%-39.7%)、4.6%(0%-20.9%)。T细胞各亚群的IL-4阳性细胞比例在动员后升高不明显。移植早期aGVHD发生组供者的Tc1细胞比例显著高于无aGVHD组。动员后高Tc2比例组,其受者中、重度aGVHD的发生率明显低于低Tc2比例组(分别为20.0%和77.8%)。结论:G-CSF动员使供者外周血的I型T细胞比例增加,动员后高Tc1细胞比例与移植早期aGVHD发生相关,Tc2比例与中重度aGVHD发生率负相关。
To investigate the effect of granulocyte colony-stimulating factor (G-CSF) mobilization on donor T-lymphocyte subsets function and acute graft-versus-host disease (aGVHD), peripheral blood samples from 20 donor G- The cells were stimulated with phorbol ester, epimycin and monensin for 4 hours in vitro. T cell subsets Ⅰ (INF-γ, INF-α) and type Ⅱ Secretion of IL-4, IL-5 and IL-10) cells. The results showed that the proportion of CD3 + IFN-γ +, CD4 + IFN-γ + and CD8 + IFN-γ + T cells in peripheral blood before mobilization was 3.2% (0% -45.9%), 1.3% % -23.8%) and 1.5% (0% -22.2%) respectively. The percentage of cells in the G-CSF group after mobilization was significantly higher than that of the G-CSF group by 19.2% (0% -53.9%) and 9.5% (0% -49.5% 7.5% (0% -38.1%). The proportions of CD3 + IL-2 +, CD4 + IL-2 + and CD8 + IL-2 + T cells before mobilization were 1.5% (0% -31%), 0.8% (0% -30.0%) and 0% (0% -39.7%), 4.6% (0% -20.9%), respectively. The proportion of IL-4 positive cells in each subset of T cells was not significantly elevated after mobilization. The proportion of Tc1 cells in the aGVHD group was significantly higher than that in the aGVHD group. After mobilization, patients with high Tc2 ratio had significantly lower rates of moderate and severe aGVHD than those with low Tc2 ratio (20.0% vs 77.8%, respectively). CONCLUSIONS: G-CSF mobilization increases the proportion of type I T cells in donor peripheral blood. After mobilization, the proportion of high Tc1 cells is correlated with the occurrence of aGVHD in early stage of transplantation. The proportion of Tc2 is negatively correlated with the incidence of moderate-severe aGVHD.
