目的 :研究人端粒重复序列结合因子 (TRF1)在 30例急性白血病细胞中的表达 ,了解 TRF1表达与急性白血病分型的关系 ,以及与端粒酶活性的关系。方法 :用荧光定量 RT- PCR定量检测 30例急性白血病细胞中TRF1m RNA表达情况 ,同时检测 h TERT m RNA的表达 ,并分析两者之间的相关性。结果 :TRF1在急性非淋巴细胞性白血病 (ANLL)细胞中的表达 0 .0 12 6 (0 .0 12 7~ 0 .0 5 46 )明显低于正常人骨髓单个核细胞中的表达 0 .0 4 5 7(0 .0 0 839~ 0 .2 6 2 ) (P<0 .0 0 1) ,但在急性淋巴细胞性白血病 (ALL)细胞中的表达 0 .0 74 5 (1.92× 10 -6~ 0 .193)与正常人骨髓单个核细胞比较差异无显著性 ,且在 ANL L细胞中的表达明显低于 ALL细胞中的表达 (P=0 .0 0 1)。TRF1和 h TERT在急性白血病细胞和正常人骨髓单个核细胞中的表达无显著相关性 (r=- 0 .173,P=0 .2 0 7)。结论 :TRF1m RNA在 ANLL细胞中表达明显降低 ,而在 ALL细胞中表达无显著改变 ,提示 ANLL细胞中 TRF1可能通过调控端粒长度对端粒酶起间接负调控作用。
Objective: To investigate the expression of TRF1 in 30 cases of acute leukemia and to investigate the relationship between the expression of TRF1 and the classification of acute leukemia and its relationship with telomerase activity. Methods: Quantitative RT-PCR was used to detect the expression of TRF1mRNA in 30 cases of acute leukemia cells. Meanwhile, the expression of h TERT m RNA was detected and the correlation between them was analyzed. Results: The expression of TRF1 in acute non-lymphocytic leukemia (ANLL) cells was significantly lower than that in normal human bone marrow mononuclear cells (P0.01 ± 0.127 ~ -0.05446) (P <0.001) but not in acute lymphoblastic leukemia (ALL) cells (P> 0.05) 6 ~ 0.193) had no significant difference with normal human bone marrow mononuclear cells, and the expression in ANL L cells was significantly lower than that in ALL cells (P = 0.010). There was no significant correlation between TRF1 and h TERT expression in acute leukemia cells and normal human bone marrow mononuclear cells (r = - 0.173, P = 0.277). CONCLUSION: TRF1mRNA was significantly decreased in ANLL cells but not in ALL cells, suggesting that TRF1 may play an indirect negative regulatory role on telomerase by regulating telomere length in ANLL cells.
