目的探讨知母总皂苷(total timosaponin,TT)对慢性温和应激模型小鼠的保护作用并分析其可能的作用机制。方法建立小鼠慢性温和应激模型,测量小鼠对蔗糖水的摄取量,采用避暗实验考察小鼠学习记忆能力,放射免疫法测定小鼠血浆促肾上腺皮质激素(ACTH)和皮质醇含量,酶联免疫法测定海马脑源性神经生长因子(BDNF)含量。结果 TT(25、50 mg.kg-1,qd×21 d)可显著增加慢性温和应激模型动物的蔗糖水消耗量,延长动物逃避潜伏时间、减少逃避错误次数。生化结果显示,TT能够降低血浆ACTH和皮质醇的含量,提高海马组织内BDNF含量。结论 TT能够改善慢性温和应激模型小鼠的抑郁状态和学习记忆能力,其机制与抑制下丘脑-垂体-肾上腺轴功能亢进以及提高模型动物海马BDNF含量有关。
Objective To investigate the protective effect of total timosaponin (TT) on chronic mild stress model mice and to analyze its possible mechanism. Methods The model of chronic mild stress was established in mice. The intake of sucrose in mice was measured. The learning and memory abilities of mice were observed by dark-avoidance test. The levels of adrenocorticotropic hormone (ACTH) and cortisol were measured by radioimmunoassay. Determination of hippocampal brain derived nerve growth factor (BDNF) by enzyme - linked immunosorbent assay. Results TT (25,50 mg.kg-1, qd × 21 d) significantly increased sucrose water consumption in chronic mild stress model animals, prolonged the escape latency and reduced the number of avoidance errors. Biochemical results show that TT can reduce plasma ACTH and cortisol levels, improve BDNF content in hippocampal tissue. Conclusion TT can improve depression and learning and memory abilities in chronic mild stress model mice, and its mechanism is related to inhibition of hypothalamic-pituitary-adrenal axis hyperactivity and elevation of BDNF content in hippocampus of model animals.
