目的:探讨白介素-6(IL-6)及基质金属蛋白酶-2(MMP-2)在胚胎发育中的可能作用。方法:对30例不明原因胚胎停止发育病例(研究组)和30例正常早孕吸宫流产病例(对照组)的绒毛组织及血清进行研究。采用实时荧光定量逆转录聚合酶链式反应技术(Real time-PCR)和免疫组织化学技术(SP法)检测两组绒毛组织中IL-6和MMP-2 mRNA的表达和蛋白定位、表达强度;采用双抗体酶联免疫夹心法(ELISA法)测定IL-6在两组血清中的浓度。结果:绒毛组织中,IL-6与MMP-2在合体及绒毛滋养层细胞中均有表达,Real time-PCR及免疫组化结果均显示IL-6在研究组中的表达水平低于对照组,MMP-2在研究组中的表达水平高于对照组,差异均有统计学意义(P<0.05);IL-6在血清中的表达水平组间无统计学差异(P>0.05)。结论:孕早期绒毛组织中IL-6和MMP-2的正常表达对维持早期胚胎发育起到一定作用。
Objective: To investigate the possible role of interleukin-6 (IL-6) and matrix metalloproteinase-2 (MMP-2) in embryonic development. Methods: The villus tissues and sera of 30 cases of unexplained embryo stop development (study group) and 30 cases of normal pregnancy induced abortion (control group) were studied. The mRNA expression and protein localization of IL-6 and MMP-2 in the two groups were detected by real time-PCR and immunohistochemistry (SP method). The concentration of IL-6 in serum of both groups was determined by enzyme-linked immunosorbent assay (ELISA). Results: The expression of IL-6 and MMP-2 in trophoblastic cells of both villi and villi was observed in villi. The results of Real time-PCR and immunohistochemistry showed that the expression of IL-6 in the study group was lower than that in the control group , While the expression of MMP-2 in the study group was higher than that in the control group (P <0.05). There was no significant difference in the expression level of IL-6 between the two groups (P> 0.05). Conclusion: The normal expression of IL-6 and MMP-2 in the first trimester of pregnancy may play an important role in maintaining early embryonic development.
