目的探讨肿瘤干细胞相关基因Bmi-1、POU5F1在子宫颈癌组织中的表达及其与临床病理特征的关系。方法采用实时荧光定量PCR(QRT-PCR)检测69例子宫颈癌组织和16例子宫颈癌组织及其配对的癌旁新鲜组织标本中Bmi-1 mRNA、POU5F1 mRNA的表达水平。结果子宫颈癌组织中Bmi-1 mRNA、POU5F1 mRNA相对表达量分别为1.51±0.05和1.75±0.17,高于配对癌旁组织的1.16±0.17和1.46±0.18,差异均有统计学意义(P<0.05)。Bmi-1 mRNA、POU5F1 mRNA在宫颈癌组织与配对癌旁组织中表达的倍比关系分别为1.31±0.49和1.20±0.95。Bmi-1 mRNA表达与民族、淋巴结转移、临床分期及组织学分级有关(P<0.05),而与年龄、肿瘤大小及病理类型无关(P>0.05)。POU5F1 mRNA表达与民族、肿瘤大小、淋巴结转移、临床分期及组织学分级有关(P<0.05),而与年龄及病理类型无关(P>0.05)。结论肿瘤干细胞相关基因Bmi-1、POU5F1在子宫颈癌组织中高表达,两者可能与子宫颈癌的不良生物学特性关系密切,可作为评估子宫颈癌生物学行为的潜在分子标志物。
Objective To investigate the expression of tumor-associated genes Bmi-1 and POU5F1 in cervical cancer and their relationship with clinicopathological features. Methods The expression of Bmi-1 mRNA and POU5F1 mRNA in 69 cases of cervical cancer tissues and 16 cases of cervical cancer tissues and matched paracancerous tissues were detected by real-time fluorescence quantitative PCR (QRT-PCR). Results The relative expressions of Bmi-1 mRNA and POU5F1 mRNA in cervical cancer tissues were 1.51 ± 0.05 and 1.75 ± 0.17, respectively, which were significantly higher than those in para-cancerous tissues (1.16 ± 0.17 and 1.46 ± 0.18, P < 0.05). The ratio of Bmi-1 mRNA and POU5F1 mRNA expression in cervical cancer tissues and adjacent tissues was 1.31 ± 0.49 and 1.20 ± 0.95, respectively. The expression of Bmi-1 mRNA was related to ethnicity, lymph node metastasis, clinical stage and histological grade (P <0.05), but not to age, tumor size and pathological type (P> 0.05). POU5F1 mRNA expression was related to ethnicity, tumor size, lymph node metastasis, clinical stage and histological grade (P <0.05), but not to age and pathological type (P> 0.05). Conclusion The expression of Bmi-1 and POU5F1 in cervical cancer tissues is high, which may be closely related to the adverse biological characteristics of cervical cancer. It may be used as a potential molecular marker to evaluate the biological behavior of cervical cancer.
